Tretinoin


Tretinoin

Class:Cytostatics,Anti-acne preparation

Mechanism of action

Keratinocytes in the sebaceous follicle become less adherent which allows for easy removal;inhibits microcomedo formation and eliminates lesions already present.

Pharmacokinetics

Absorption:Topical:minimum absorption occur.Metabolism:of the small amount absorbed,metabolism occur in the liver.elimination in the bile and urine.

Indications

Topical:acne vulgaris.oral:with anthracycline based chemotherapy in acute promyelocytic leukemia.

Contraindications

Acute eczema,rosacea and acute inflammation of the skin.

Precautions

Contact of tretinoin with eyes,anterior naris and mucosa should be avoided;otherwise thorough rinsing with water is recommended.

Pregnancy and lactation

Should be avoided in pregnancy

Drug interaction

Special caution is indicated in case of simultaneous treatment with other preparations,especially desquamative agents.if the patients was treated with such an agent prior to the tretinoin therapy,it is recommended that the skin be allowed to recover.

Dosage

Cream shall be applied once or twice daily for minimum 8 weeks.Oral:adults and children:45mg/square meter daily in two divided doses with a meal.contnue treatment until complete remission or up to maximum 90 days.
