Mechanism of action
Antiplasminic binds
strongly with the lysine-bonding site(LBS),the site of fibrin affinity of
plasmin and plasminogen,and inhibits the bonding of plasmin and plasminogen to
fibrin.Therefore,the degradation of fibrin by plasmin is strongly inhibited.In
the presence of antiplasmins,such as u2-macro globulin,in the plasma,the
antifibrinolytic action of tranexamic acid is even further strenghthened.In
case of ordinary haemorrhages,Tranexamic acid appear to cause hematosis by
suppressing this fibrin degradation.
Pharmacokinetics
The concentrations in
the blood,when 250mg and 500mg of tranexamic acid were administered orally to
healthy adults,reached peak values 3.9 ug/ml(250mg administration) and 6.0
ug/ml (500mg administration) 2-3 hours after administration.The biological half
lives was 3.1 hours and 3.3 hours,respectively.When 500mg of tranexamic acid
were administered orally to healthy adults,the rate of excretion in the urine was
about 30-52% by 24 hours after administration.
Indications
Leukemia,hypoplastic
anemia,purpura or abnormal hemorrhages during or after
operation.pulmonary,nasal,genital or renal hemorrhages,eczema or similar
diseases,urticaria,drug rash or toxic
exanthema.Tonsillitis,pharyngolaryngitis,pain in the oral cavity or mucosal
aphtha in cases of stomatitis.
Side effects
Hypersensitivity,anorexia,nausea,vomiting,diarrhea,constipation,itching,exanthema,drowsiness.

Drug interactions
Since there is a
possibility of thrombus formation tendencies due to concomitant administration
with large doses of hemostatic organ preparation or hemocoagulate,is careful
when administering concomitantly.
Dosage
Adults:The dosage of
tranexamic acid is 750 to 2000mg/day of tranexamic acid is usually administered
three to four times daily.The dosage should be adjusted according to the age of
patients and severity of symptoms.
