Orlistat

Class:Lipase inhibitor

Mechanism of action

Orlistat is a potent,specific and reversible long acting inhibitor of gastrointestinal lipases.It exert its therapeutic activity in the lumen of the stomach and small intestine by forming covalent bond with the serine residue of the active site of gastric and pancreatic lipases.The inactivated enzyme is thus unable to hydrolyse dietary fat,in the form of triglycerides,into absorbable free fatty acids and monoglycerides.As undigested triglycerides are not absorbed.

Indications

Obese patients or severly overweight patients in conjunction with a mildly hypocaloric diet.

Contraindications

Chronic malabsorption syndrome or cholestasis,hypersensitivity to orlistat or any component.

Side effects
Headache,dizziness,sleep disorder,anxiety,dry skin,rash,oily spotting,fatty/oily stool.

Pregnancy

Pregnancy risk factor B.

Drug interaction

Vitamin K absorption may be decreased when taken with orlistat.

Dosage

One capsule of 120mg during or upto one hour after the meal.
